How It Works
Polynucleotides are higher-molecular-weight DNA polymers than PDRN — longer chains that provide both a direct regenerative stimulus and a biopolymer scaffold within the dermis. They activate fibroblasts via adenosine receptor pathways (primarily A2A and A1) to produce new collagen type I, type III, and elastin fibres. Their hydrophilic nature also directly attracts and retains water within the dermis, improving deep hydration through a mechanism complementary to — but distinct from — hyaluronic acid. Ultrasound imaging in clinical studies has shown measurable increases in dermal thickness following PN treatment.
Clinical Evidence
PN skin boosters are the fastest-growing category in aesthetic medicine. A 2021 prospective clinical study found that PN injections produced a significant increase in skin hydration (corneometry), with 82% of participants rating improvement at 8 weeks. A 2022 pilot study found a 23% increase in ultrasound-measured dermal thickness after 8 weeks of topical 1.5% PN application. A 2023 RCT showed a 19% reduction in transepidermal water loss in subjects using a PN-based topical formulation, indicating improved barrier function.

How to Use
Use PN serum morning and evening after cleansing and toning. The ingredient works cumulatively — expect visible changes in skin texture and plumpness after 4–6 weeks of consistent use. Layer under a ceramide-rich moisturiser to amplify barrier support. When used alongside injectable PN treatments, topical PN helps maintain and extend clinical results between sessions.
